There are two very different roads to a broken protein, and Stina’s case runs down both. Pyrin misfolds because of an inherited typo, present from the start; amyloid aggregates slowly, building up over years of inflammation no one was treating. And late in her record comes the most validating sentence she has ever read about herself — “persistently elevated acute-phase reactants.” Translated out of clinical shorthand, it means the high markers that had haunted even her good weeks were real, measurable, and ongoing the entire time. That single line is Chart Clue #2: the moment her bloodwork stopped being explained away and started being believed.

A protein’s amino-acid sequence dictates its folded shape, and when folding fails, function is lost. Stina’s case shows both failure modes side by side. Pyrin carries an inherited misfold, written into the gene itself, so the protein is built wrong from the first copy. Amyloid is the acquired route: ordinary proteins, over years, stacking into stubborn, insoluble beta-sheets that lodge in organs and refuse to leave. One error is in the blueprint; the other accumulates from wear. Both end in a protein that no longer does its job.
